Cytokine and growth factor pathways

Overview

Cytokine and growth factor pathways represent a broad and complex network of signaling cascades that govern essential cellular processes including growth, differentiation, and immune regulation (NIH, 2023). Cytokines, such as interleukins and interferons, primarily coordinate the immune system's response to infection and injury, while growth factors like VEGF and EGF regulate tissue repair and development (StatPearls, 2023). These pathways typically involve the binding of a secreted ligand to a specific cell-surface receptor, which then activates intracellular signaling mediators like the JAK/STAT or MAPK pathways to alter gene expression (UniProt, 2024). Dysregulation of these pathways is a primary driver in various diseases; for instance, pro-inflammatory cytokine overproduction leads to chronic inflammatory disorders, while aberrant growth factor signaling is frequently implicated in oncogenesis and metastasis (PubMed, 2022). Consequently, these pathways are major focuses of drug development, with therapies ranging from monoclonal antibodies that neutralize specific cytokines to small-molecule inhibitors targeting downstream kinases (Nature Reviews Drug Discovery, 2021). Because this term encompasses hundreds of distinct proteins and interactions, it is classified as a pathway category rather than a single therapeutic target. Targeting these pathways requires high specificity to avoid significant off-target effects and systemic toxicity. Clinical monitoring often involves measuring specific cytokine levels or receptor expression to tailor therapy to the patient's molecular profile.

Mechanism of action

Therapeutic intervention involves the use of monoclonal antibodies to neutralize ligands or block receptor binding, as well as small-molecule inhibitors to disrupt intracellular signaling components like tyrosine kinases (PubMed, 2023).
