Cytokine-dependent hematopoietic cell linker (CLNK) is a member of the SLP-76 family of SH2-domain–containing adaptor proteins[1][5][9][12]. CLNK is involved in the regulation of immunoreceptor signaling across multiple lymphocyte lineages, including B cells, T cells, natural killer (NK) cells, and mast cells[1][5][9][12]. In B and T cells, it helps coordinate proximal receptor signaling with downstream cascades such as PLC-gamma activation, influencing cell activation, cytokine production, and, in some contexts, cell death pathways such as necroptosis[1][5][12]. While initially implicated in mast cell signaling (thus the alias MIST, "mast cell immunoreceptor signal transducer"), functional studies indicate its role in mast cell degranulation may be redundant or auxiliary[1]. Further, CLNK acts as a regulator (positive or negative depending on the cell context) of T- and NK-cell activation and can modulate immune responses and exhaustion[1][5][12]. Elevated serum CLNK has been observed in transfusion-dependent thalassemia and correlates with iron overload, suggesting potential as a biomarker in certain contexts[7]. There are currently no approved therapeutics targeting CLNK or drugs known to interact specifically with this protein.
