Overview

Cytokine-inducible SH2-containing protein (CISH) is a member of the suppressor of cytokine signaling (SOCS) family. It acts as an inducible inhibitor of cytokine signaling, negatively regulating T cell receptor (TCR) signaling and natural killer (NK) cell cytotoxicity. Dysregulation of CISH is linked to various diseases, including infections, asthma, anemia, and cancer. Targeting CISH enhances the efficacy of adoptive T-cell and NK cell therapies against tumors.

Mechanism of action

Inhibition of CISH enhances T cell and NK cell activity against tumors. Targeting CISH in NK cells improves ex vivo proliferation and limits NK cell exhaustion, impairing primary tumor and metastasis development.
