Cytokine-mediated antitumor activity

Overview

Cytokine-mediated antitumor activity refers to the suppression or elimination of tumor cells by modulating immune system responses through cytokines—small glycoproteins that regulate immune cell activation, proliferation, differentiation, and apoptosis[1][3][6][7]. This process can involve direct inhibition of tumor growth and promotion of tumor cell apoptosis, or indirect effects such as activating T cells, natural killer cells, and antigen-presenting cells to attack tumors. Key cytokines with documented antitumor activities include interleukin-2, interferon alpha, interferon gamma, and GM-CSF, among others[1][3][6][7]. Therapeutic strategies targeting cytokine pathways include recombinant cytokines, monoclonal antibodies, receptor antagonists, and engineered cytokine variants. Major challenges include systemic toxicities, limited efficacy as monotherapy, and potential for promoting pro-tumor immune suppression or evasion[3][6].

Mechanism of action

Stimulation of effector T cell and NK cell activity, promotion of tumor antigen presentation and T-cell priming, induction of apoptosis in tumor cells, inhibition of regulatory immune cells within the tumor microenvironment, and modulation of cytokine-receptor interactions.
