Overview

The cytokine receptor common subunit beta, also known as CSF2RB or CD131, is a critical cell surface receptor component involved in immune regulation and hematopoiesis. It serves as the shared signaling subunit for the receptors of three type I cytokines: granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-3 (IL-3), and interleukin-5 (IL-5). Upon ligand binding to the receptor complex, CSF2RB initiates intracellular signaling cascades—primarily through JAK1 activation leading to downstream JAK–STAT pathway signaling—which regulate immune responses and control production/differentiation of hematopoietic progenitor cells into various blood lineages. Alterations in βc-mediated pathways have been implicated in various diseases including chronic inflammation and malignancies.
