Overview

Cytokine receptor-like factor 1 (CRLF1) is a secreted protein belonging to the cytokine type I receptor family and is structurally related to receptors in the interleukin-6/gp130 cytokine family[1][2][3][4]. CRLF1 forms either a homodimer or a heterodimer with cardiotrophin-like cytokine factor 1 (CLCF1/CLC)[1][2][3][4][5]. The heterodimeric CRLF1/CLC complex signals via the ciliary neurotrophic factor receptor (CNTFR), gp130, and leukemia inhibitory factor receptor (LIFR), promoting STAT3 activation and cell survival, especially in neuronal cells[1][4][5]. Homodimeric CRLF1 signals independently and is linked to neuroprotection and tissue regeneration, but its precise receptor is unknown[1][5]. Mutations in CRLF1 result in rare syndromes such as cold-induced sweating syndrome and Crisponi syndrome, characterized by autonomic dysfunction and craniofacial anomalies[3][4]. In disease, CRLF1 is upregulated in osteoarthritic cartilage, and promotes fibrotic tissue remodeling via ERK signaling, especially in ligamentum flavum hypertrophy, making it a potential biomarker and therapeutic target for fibrotic and degenerative diseases[2][1].
