The Interleukin-6 receptor (IL-6R) and Interleukin-10 receptor (IL-10R) are members of the Type I and Type II cytokine receptor families, respectively, and primarily utilize the Janus kinase-signal transducer and activator of transcription (JAK-STAT) pathway to regulate gene expression [PMID: 29080852]. The Interleukin-8 receptors (CXCR1 and CXCR2) are G protein-coupled receptors (GPCRs) that drive the recruitment and activation of neutrophils to sites of injury or infection [PMID: 30213465]. The Interferon-gamma receptor (IFNGR) is essential for activating macrophages and promoting Th1-type immune responses, making it a key player in antiviral and antitumor defense [PMID: 10359101]. Dysregulation of these receptor systems is a hallmark of various pathologies, including rheumatoid arthritis, cytokine release syndrome (CRS), and chronic inflammatory disorders. Consequently, they are major therapeutic targets for monoclonal antibodies, such as tocilizumab for IL-6R and emapalumab for IFN-gamma signaling, as well as small molecule inhibitors targeting their downstream signaling cascades [PMID: 32433942].
