Overview

The Cytomegalovirus, Epstein-Barr virus, and Influenza virus (CEF) peptide pool is a standardized collection of immunodominant peptides derived from these three common human viruses (Mabtech, 2023). It is primarily utilized as a positive control in immunological assays, such as ELISpot and flow cytometry, to stimulate antigen-specific CD8+ T cells in individuals with common HLA types, particularly HLA-A*0201 (Currier et al., 2002). When these peptides are presented by Human Leukocyte Antigen (HLA) molecules on the surface of dendritic cells or other antigen-presenting cells, they mimic viral infection and trigger a robust memory T-cell response (JPT Peptide Technologies, 2023). This response is characterized by the production of effector cytokines like interferon-gamma, which are measured to assess the functional integrity of a patient's cellular immune system. While the CEF pool is not a therapeutic target itself, it is an essential tool for validating the efficacy of vaccines and immunotherapies by providing a reliable benchmark for T-cell reactivity. It is widely employed in clinical research to monitor the baseline and post-treatment immune competence of patients undergoing various treatments.

Mechanism of action

The CEF peptide pool functions by providing immunodominant viral epitopes that are loaded onto MHC Class I molecules of antigen-presenting cells (APCs). These complexes are recognized by the T-cell receptors (TCRs) of pre-existing memory CD8+ T cells, inducing cellular activation, proliferation, and the secretion of pro-inflammatory cytokines.
