Cytomegalovirus (CMV) DNA polymerase is a viral enzyme essential for the replication of CMV. It is the primary target for several antiviral drugs used to treat CMV infections, particularly in immunocompromised patients. The enzyme consists of two main subunits: the catalytic subunit (UL54) and the accessory subunit (UL44). UL54 is responsible for DNA synthesis, while UL44 acts as a processivity factor. The CMV DNA polymerase is inhibited by antiviral agents like ganciclovir, foscarnet, and cidofovir, which are approved treatments against severe CMV disease. Resistance can develop through mutations primarily within functional regions under selective pressure from antiviral therapy.
