The cytomegalovirus DNA terminase complex subunit pUL56 is a key protein involved in the replication cycle of human cytomegalovirus (HCMV). It is a major component of the viral terminase complex, essential for packaging viral DNA into capsids during virion assembly. pUL56 recognizes and binds specific packaging motifs on viral DNA, acts as an "anchor" for other terminase subunits, facilitates translocation of viral DNA into preformed capsids, exhibits ATP-independent endonuclease activity, and provides ATP necessary for genome encapsidation. It is the primary target of the antiviral drug letermovir.
