pUL56 is a key subunit of the human cytomegalovirus (HCMV) terminase complex, essential for viral DNA maturation and packaging. It functions as an ATPase and DNA-binding protein, anchoring the terminase complex to viral DNA and providing energy for DNA translocation into capsids. It interacts directly with pUL89 and pUL104. As its structure is unique to herpesviruses, pUL56 represents an attractive target for antiviral therapy; the drug letermovir inhibits HCMV replication by targeting this protein. Resistance mutations have been mapped to the UL51, UL56, and UL89 genes.
