Cytoplasmic differentiation-associated epidermal RNA (CYDAER), also known as RP11-295G20.2 and TSNAX-DT, is a skin-enriched long non-coding RNA predominantly found in the cytoplasm of suprabasal layers of the epidermis. CYDAER is dynamically regulated during keratinocyte differentiation, being upregulated in early differentiation and downregulated during late stages. In psoriasis, CYDAER is strongly induced by IL-17A and its overexpression may contribute to abnormal epidermal differentiation. In hepatocellular carcinoma, it is significantly upregulated and acts as an oncogenic lncRNA by promoting cancer cell proliferation and inhibiting autophagy. Mechanistically, CYDAER binds directly to the tumor suppressor protein PTEN, leading to its degradation via the lysosomal pathway, which results in downstream activation of AKT signaling and repression of autophagy-related genes. This lncRNA functions in both epidermal biology and cancer, but there are no data supporting a direct role as a classical drug target or receptor.
