Overview

Cytoskeleton-associated protein 2 (CKAP2) is a microtubule-associated protein that plays a crucial role in mitotic spindle assembly and the regulation of microtubule growth dynamics during cell division. It acts as a microtubule nucleation and growth factor, stabilizing microtubules and suppressing catastrophes (sudden depolymerizations), making it one of the most potent microtubule growth factors characterized to date[1][2][4][5]. CKAP2 localizes to centrosomes and spindle fibers during mitosis, ensuring accurate chromosome segregation; upon mitotic exit, it is rapidly degraded. Loss of CKAP2 disrupts microtubule dynamics and increases chromosomal missegregation and aneuploidy. Misregulation—especially overexpression—of CKAP2 is linked to tumorigenesis and poorer survival in several cancers, with the gene also showing high activity in proliferative tissues and lymphomas[1][3][7]. CKAP2 is regulated by microRNAs and post-translational phosphorylation, and its gene resides in a chromosome region prone to deletions and rearrangements in various tumors[3][7]. There are currently no known direct small-molecule or antibody drugs that specifically target CKAP2; its overexpression, however, may serve as a biomarker for cancer cell proliferation and aggressiveness[1][5][7].
