Cytoskeleton-associated protein 5 (CKAP5), also known as ch-TOG and a member of the TOG/XMAP215 family, is a highly conserved microtubule-associated protein. CKAP5 is a processive microtubule polymerase that binds tubulin and the plus-ends of microtubules, promoting microtubule nucleation, elongation, and overall cytoskeletal organization[1][3][7]. It plays essential roles in mitosis, protecting kinetochore microtubules from depolymerization and facilitating spindle pole integrity and bipolar spindle formation. CKAP5 also contributes to cell movement through its regulation of microtubule-cortex interactions and is involved in neuron outgrowth via its unique ability to cross-link microtubules and F-actin[5]. CKAP5 acts as a core component of the TACC3/ch-TOG/clathrin complex, which is critical for stabilization of kinetochore fibers and the tension of the mitotic spindle[1][3][2]. Although not a classic therapeutic target like receptors or enzymes, CKAP5 plays important roles in cell proliferation and has been implicated in cancer and neurodegenerative diseases[3][7]. No approved drugs are known to target CKAP5 directly.
