Overview

Cytosolic carboxypeptidase 6 (AGBL4) is a metallocarboxypeptidase enzyme that catalyzes the deglutamylation of polyglutamate side chains on the C-terminal tails of tubulin and other proteins[4][3]. This modification is essential for regulating microtubule function and protein stability. AGBL4 is implicated in various biological pathways, including axonal transport, protein catabolism, blastocyst development, and antiviral defense response[3][2]. In cancer, notably glioblastoma, AGBL4 promotes malignant cell proliferation, migration, and invasion, and its overexpression correlates with poorer prognosis, possibly through upregulation of matrix metalloproteinase-1 (MMP-1) and interaction with inflammatory pathways[1]. Epigenetic silencing and genetic variants of AGBL4 have been linked to disease risk, metabolic traits, and serve as potential biomarkers for early detection and disease monitoring in colorectal cancer and cardiometabolic disorders[2]. The enzyme is primarily localized to the cytosol, Golgi apparatus, centriole, and ciliary basal body[3]. Currently, there are no drugs known to specifically target AGBL4, nor are there characterized mechanisms of action or safety concerns for therapeutic intervention in clinical use.
