Cytosolic iron-sulfur assembly component 3 (CIAO3) is a protein that is part of the cytosolic iron-sulfur cluster assembly complex (CIA complex), which is essential for the incorporation of iron–sulfur ([4Fe-4S]) clusters into extramitochondrial Fe-S proteins[8][5][3]. CIAO3 forms stable complexes with other CIA machinery proteins such as CIAO1 and CIA2A, orchestrating the late steps in cytosolic and nuclear Fe–S protein maturation[1]. It participates in oxygen homeostasis and the cellular response to hypoxia and appears to negatively regulate the expression of HIF1A, possibly through its iron-sulfur cluster assembly function[3][7][8]. Defects in iron–sulfur cluster assembly factors like CIAO3 can contribute to rare diseases such as Fanconi anemia and may compromise essential cellular processes, but CIAO3 itself is not currently considered a direct therapeutic target for drugs[3][8].
