Overview

Cytosolic proteins represent a vast and heterogeneous collection of proteins located within the cytosol, the aqueous component of the cytoplasm excluding organelles. This group includes essential enzymes involved in metabolic pathways like glycolysis, structural proteins that maintain the cytoskeleton, and critical signaling molecules such as kinases and phosphatases (Alberts et al., 2014, Molecular Biology of the Cell). Because many of these proteins are central to cellular survival, growth, and communication, they are frequent focal points for drug discovery in oncology, immunology, and metabolic diseases (Hopkins & Groom, 2002, Nature Reviews Drug Discovery). However, the term "Cytosolic proteins" is a broad classification based on subcellular localization rather than a specific, discrete therapeutic target. In clinical practice and drug development, specific members of this class, such as BCR-ABL or the proteasome, are targeted to achieve therapeutic effects. Effective modulation of these proteins often requires small molecules capable of crossing the plasma membrane or advanced technologies like Proteolysis-Targeting Chimeras (PROTACs) to reach the intracellular environment (Sun et al., 2019, Signal Transduction and Targeted Therapy).

Drugs targeting specific cytosolic proteins typically function through competitive or non-competitive inhibition of enzymatic activity, modulation of protein-protein interactions, or the induction of targeted protein degradation via the ubiquitin-proteasome system (Sun et al., 2019, Signal Transduction and Targeted Therapy).
