Cytotoxic activity via HLA-restricted recognition of viral antigens refers to the process by which cytotoxic T lymphocytes recognize viral peptides that are presented on the surface of infected cells by human leukocyte antigen (HLA, also known as MHC in humans) molecules. This recognition is highly specific—T cell receptors on cytotoxic T cells bind to complexes of particular HLA alleles (MHC class I for CD8^+^ T cells, class II for CD4^+^ T cells) with viral peptides. Once recognition occurs, the cytotoxic T cell is activated to kill the infected cell, thereby contributing to the immune clearance of the virus. This process is fundamental to adaptive immunity, crucial in infections, cancer surveillance, and autoimmunity.
Therapeutic interventions modulate components of this process, for example, via enhancement of T cell cytotoxic activity (e.g., cytokine stimulation), checkpoint inhibition (e.g., PD-1/PD-L1 blockade), or vaccine-mediated induction of specific cytotoxic T cells.
