Cytotoxic payload release

01

Overview

“Cytotoxic payload release” is not a specific molecule, receptor, or canonical therapeutic target. Instead, it describes the process by which cytotoxic agents (payloads) are liberated from carrier molecules—most commonly in the context of antibody–drug conjugates (ADCs)—after internalization into target cells such as cancer cells. In ADC therapy, an antibody binds to an antigen on the tumor cell surface and is internalized; lysosomal degradation then releases the attached cytotoxic payload inside the cell, leading to cell death[1][3]. The efficiency and properties of this intracellular release are critical for therapeutic efficacy and safety. The term does not refer to any single protein or gene but rather a pharmacological mechanism central to ADC function. Therefore, “cytotoxic payload release” should not be treated as a molecular drug target but as an essential step in certain drug delivery strategies[1][2].

02

Mechanism of action

Not applicable to a molecular target; refers to the release of cytotoxic agents from antibody-drug conjugates within cells[1][2][3].

03

Biological functions

Other (as a process, not a molecule)
04

Disease associations

Cancer (in the context of antibody-drug conjugates and targeted therapies)
05

Safety considerations

Systemic toxicity due to off-target or bystander effects if payload is released outside intended cells[1].Potential for healthy tissue damage if membrane-permeable payloads diffuse beyond target tumor cells[1][2].
