Overview

Cytotoxic T lymphocyte (CTL) activation via HLA-mediated antigen presentation is a central process in adaptive immunity. It involves the recognition of peptide antigens presented by HLA class I molecules on the surface of cells by CD8+ T cells, leading to CTL activation and killing of target cells. This mechanism is crucial for eliminating infected or malignant cells. The process involves intracellular protein degradation, peptide transport into the ER, HLA class I molecule binding, cell surface presentation, and CTL recognition via TCRs. All nucleated human cells express HLA class I molecules, enabling immune surveillance. The pathway is essential for detecting and eliminating virus-infected or transformed host cells, with defects leading to increased susceptibility to infections or cancer. It is also central to transplant rejection due to HLA differences between donor and recipient.
