Overview

Cytotoxic T lymphocyte (CTL) activation via major histocompatibility complex (MHC) presentation refers to the process by which CD8+ cytotoxic T cells are activated upon recognizing antigens presented by MHC class I molecules on the surface of target cells. This mechanism is central to adaptive immunity, enabling the immune system to identify and eliminate infected or malignant cells. The process involves antigen recognition via the T cell receptor (TCR) interacting with peptide-MHC class I complexes, co-stimulation through molecules like CD28 and CD80/86, intracellular signaling cascades leading to CTL activation, and effector functions such as perforin/granzyme release and Fas/FasL-mediated apoptosis. Dysregulation of this pathway can lead to immunodeficiency or autoimmunity. Therapeutic interventions targeting this pathway include checkpoint inhibitors, vaccines, and immunosuppressants.
