Overview

Cytotoxic T lymphocytes (CTLs), also known as CD8+ T cells, are a subset of T cells that play a central role in the immune system’s ability to recognize and eliminate infected or malignant cells. Their activation is critically dependent on the presentation of tumor-associated antigens (TAAs) by major histocompatibility complex class I (MHC I) molecules on target cell surfaces. This process involves antigen processing, MHC I presentation, TCR recognition, co-stimulation, and effector functions like granule release and FasL/Fas interaction leading to target cell apoptosis. Tumors can evade this mechanism through various strategies, including downregulation of MHC I expression. Therapies aimed at enhancing this process, such as checkpoint inhibitors and adoptive cell therapies, are crucial in cancer immunotherapy.
