Cytotoxic T lymphocyte associated antigen 4 (CTLA-4), also known as CD152, is a critical immune checkpoint receptor that plays a central role in regulating T cell-mediated immune responses and maintaining self-tolerance. It acts as a negative regulator of immune activation by binding to CD80 (B7-1) and CD86 (B7-2) on antigen-presenting cells, inhibiting further T cell activation.
Blocking antibodies enhance antitumor immunity by promoting effector function while depleting intratumoral regulatory populations via mechanisms including antibody-dependent cellular cytotoxicity (ADCC).
