D-Ala-D-Ala ligase (Ddl) is an essential bacterial enzyme that catalyzes the ATP-dependent condensation of two D-alanine molecules to form the dipeptide D-alanyl-D-alanine. This dipeptide is a critical building block for peptidoglycan, a vital component of the bacterial cell wall. Due to its essential role in bacteria but absence from humans, this enzyme is a validated target for antibiotic development. Inhibiting this enzyme disrupts cell wall synthesis leading to bacterial death.
