D-alanyl-D-alanine synthetase (D-alanine--D-alanine ligase) is an essential bacterial enzyme that catalyzes the ATP-dependent formation of the dipeptide D-Ala–D-Ala from two molecules of D-alanine. This reaction is a critical step in peptidoglycan biosynthesis, which is fundamental for bacterial cell wall integrity and survival. Because of its universal and essential function in bacteria—and absence in humans—it represents an attractive target for antibiotic development.
Inhibition of peptidoglycan synthesis, leading to bactericidal effects. D-cycloserine acts via formation of a phosphorylated adduct within the active site that mimics reaction intermediates and blocks enzymatic activity.
