The D4Z4 region refers to a macrosatellite repeat array located on chromosome 4q35 (and also on 10q), composed of tandemly repeated units of approximately 3.3 kb each. The methylation status of CpG sites within these repeats is crucial for the regulation of gene expression, particularly the *DUX4* gene, which is implicated in facioscapulohumeral muscular dystrophy (FSHD). Restoring normal levels or patterns of DNA methylation at the D4Z4 region—especially at key regulatory sites—represents a therapeutic strategy aimed at re-establishing repression of aberrant *DUX4* expression.
