Daboia russelii siamensis snake venom serine proteases (SVSPs) are a group of enzymes found in the venom of the Eastern Russell's Viper that significantly impact the host's coagulation system. The most prominent member of this family is the Factor V activator (RVV-V), a highly specific serine protease that converts Factor V into its active form, Factor Va, thereby promoting the formation of the prothrombinase complex (UniProt, 2024). This enzymatic activity leads to rapid thrombin generation, resulting in venom-induced consumptive coagulopathy (VICC), which is a hallmark of Russell's viper envenomation (PubMed, 2022). These proteases are the primary targets for therapeutic intervention using antivenoms, which contain antibodies designed to bind and neutralize the venom's toxic components (WHO, 2016). Beyond their role in envenomation, SVSPs like RVV-V are utilized in clinical laboratories as diagnostic tools, specifically in the Dilute Russell's Viper Venom Time (dRVVT) test to detect lupus anticoagulants (StatPearls, 2023). Research into small-molecule inhibitors, such as nafamostat mesylate, has also explored these enzymes as targets to provide adjunctive treatment for snakebite (PubMed, 2021). The structural integrity of these enzymes relies on a conserved catalytic triad, making them susceptible to various serine protease inhibitors.
