Dachshund family transcription factor 2 (DACH2) is a protein-coding gene and a vertebrate homolog of the Drosophila dachshund gene, encoding a transcription factor involved in cell fate determination during development of organs such as the eye, limb, and genital disc. The DACH2 protein contains two characteristic domains: an N-terminal DNA-binding domain and a C-terminal domain involved in protein-protein interactions. DACH2 is expressed in embryonic neural tissues, sensory organs, muscles, and developing limbs, indicating its broad developmental roles. In muscle, DACH2 acts as a negative regulator of myogenesis, modulating myogenin activity through mechanisms involving histone deacetylases. Expression and mutations of DACH2 have been implicated in premature ovarian failure and other congenital syndromes, and high DACH2 expression has been identified as an independent biomarker of poor prognosis in serous ovarian carcinoma, correlating with indicators of cell proliferation and DNA repair capacity. There are currently no drugs directly targeting DACH2, and its primary roles are developmental and regulatory rather than pharmacological.
