Overview

DALR anticodon-binding domain-containing protein 3 (DALRD3) is a human protein encoded by the DALRD3 gene. It contains a DALR anticodon-binding domain homologous to that in arginyl tRNA synthetases. DALRD3 forms a complex with METTL2A/B methyltransferases to specifically recognize certain arginine tRNA isoacceptors and facilitate their modification by 3-methylcytosine (m3C), an essential step for proper tRNA function. Loss-of-function mutations or reduced expression of DALRD3 cause a deficiency in m3C modification, which has been linked to severe neurological diseases, such as developmental and epileptic encephalopathy. DALRD3 is also implicated in processes related to spermatogenesis due to its gene cluster organization and preferential testis expression, potentially impacting the regulation of miRNA pathways during development. Additional data suggest DALRD3 may modulate neural pathways associated with sleep and appetite, based on altered expression in relevant brain regions. There are no known drugs, clinical biomarkers, or direct therapeutic associations with DALRD3, nor safety concerns identified in the context of therapeutic targeting[1][3][4].
