Overview

DAN domain BMP antagonist family member 5 (DAND5) is a secreted, cysteine knot-containing protein that functions as a bone morphogenetic protein (BMP) antagonist and also antagonizes NODAL signaling[1]. It plays a critical role during embryonic development by regulating organogenesis, tissue differentiation, and proper left-right body axis patterning[1][2]. DAND5 binds directly to BMP and NODAL proteins, inhibiting their signaling cascades, and is necessary for correct specification of left-right asymmetry in vertebrates[1][2]. Alterations in DAND5 function or expression can lead to congenital disorders such as visceral heterotaxy[1][2]. DAND5 gene fusions have also been observed in certain cancer types, but there are no currently known direct drug interactions or established biomarker roles[3].
