Overview

DC-STAMP domain-containing 1-antisense RNA 1 (DCST1-AS1) is a long non-coding RNA significantly upregulated in several cancers, including triple-negative breast cancer, endometrial cancer, and colorectal cancer[1][2][3][5]. It functions as an oncogenic lncRNA by promoting cell proliferation, migration, and invasion and by suppressing apoptosis in cancer cells[1][3][5]. Mechanistically, DCST1-AS1 modulates gene expression post-transcriptionally by acting as a competing endogenous RNA (ceRNA) that sponges specific miRNAs (e.g., miR-665, miR-873-5p, miR-582-5p), thereby regulating downstream targets such as HOXB5, CADM1, HMGB1, and IGF2BP1[1][3][5]. In triple-negative breast cancer, DCST1-AS1 interacts with ANXA1 to enhance TGF-β-induced epithelial–mesenchymal transition (EMT) and increases cellular resistance to chemotherapy agents such as doxorubicin and paclitaxel[2]. Its expression correlates with poor prognosis in cancer patients, and it is under investigation as both a prognostic biomarker and a potential therapeutic target[1][2][3][5].
