Overview

DDB1- and CUL4-associated factor 8-like protein 2 (DCAF8L2) is a WD40-repeat-containing protein thought to act as a substrate receptor within the CUL4-DDB1 E3 ubiquitin ligase complex[2][3][5]. As with other DCAF-type proteins, it likely mediates the recruitment of specific substrate proteins for ubiquitin-dependent proteolysis, influencing cellular processes such as chromatin modification, DNA repair, transcription regulation, cell cycle progression, and protein turnover[2][5]. DCAF8L2 is predicted to localize to the nucleus and cytoplasm and is a member of a family of WD40 proteins known for their role in assembling multi-protein complexes, particularly those involved in ubiquitin-mediated cellular regulation[3][5]. While related DCAF proteins and the broader CRL4 complex have mechanistic links to cancer, neurodevelopmental disease, and chromatin regulation, there is currently limited direct evidence that DCAF8L2 itself is a therapeutic target or that any approved drugs interact with it[3][5].
