Overview

DDB1 and CUL4 associated factor 6 (DCAF6), also known as nuclear receptor interaction protein (NRIP) and IQWD1, is a ligand-dependent nuclear receptor coactivator that interacts with steroid hormone receptors, primarily the androgen receptor and glucocorticoid receptor, enhancing their transcriptional activity. As a substrate receptor for the CUL4-DDB1 E3 ubiquitin ligase complex, DCAF6 participates in the regulation of protein stability—including stabilizing androgen receptor protein in prostate cells—and modulates processes such as cell proliferation and viral gene expression. DCAF6 has been implicated in prostate and cervical cancer biology, where its knockdown impairs cancer cell proliferation, and may also regulate human papillomavirus (HPV) gene expression through interactions that promote viral protein stability[1][2][3][6].
