Overview

De-etiolated-1 homolog (DET1) is a nuclear chromatin-binding protein functioning as a key substrate adaptor in multi-protein E3 ubiquitin ligase complexes that contain CUL4, DDB1, DDA1, and COP1. DET1 specifically recruits certain E2 enzymes and the COP1 E3 ligase, coordinating the polyubiquitination and subsequent proteasomal degradation of various transcription factors involved in key biological processes such as immune, metabolic, and neurodevelopmental regulation. DET1's structural dynamics, modulated by protein-protein interactions and post-translational modifications, govern its ability to assemble complexes that regulate cell fate and environmental response. Although critical in developmental programming and disease progression, DET1 is not currently a direct therapeutic target; rather, its modulatory role in transcription factor stability makes it a node of biological interest in cancer, neurodevelopment, and possibly resistance phenomena.
