Overview

Lipid biosynthesis is a fundamental metabolic process responsible for the de novo synthesis of lipid molecules, including fatty acids, cholesterol, and phospholipids, from simple precursors like acetyl-CoA (StatPearls, Lipogenesis, 2023). This pathway is essential for building cellular membranes, storing energy in the form of triacylglycerols, and generating vital signaling molecules such as steroid hormones and eicosanoids (NCBI, Lipid Metabolism). In various disease states, particularly metabolic syndrome, obesity, and cardiovascular disease, lipid biosynthesis is often pathologically upregulated, leading to lipid accumulation and systemic inflammation (Nature Reviews Drug Discovery, 2020). Furthermore, many cancers exploit de novo lipogenesis to support rapid membrane production and meet high energy demands during proliferation (PubMed, PMID: 27157135). Pharmacological intervention typically focuses on inhibiting specific rate-limiting enzymes within this broad pathway, such as HMG-CoA reductase (targeted by statins) or Acetyl-CoA carboxylase, to manage cholesterol levels and metabolic health (NIH, Lipid Biosynthesis).
