Overview

DEAD-box helicase 4 (DDX4, also known as VASA) is a member of the DEAD-box family of ATP-dependent RNA helicases, named for its conserved amino acid motif Asp-Glu-Ala-Asp. These helicases are critical for a range of RNA metabolic processes, including transcription, RNA splicing, ribosome assembly, RNA transport, translation, and degradation[1][2][3]. DDX4 is particularly notable for its crucial role in germ cell development and is a well-established marker for germ cells. Members of this family regulate gene expression, cell cycle progression, and are implicated in cancer initiation, progression, and metastasis, making them both therapeutic targets and candidate biomarkers, especially within oncology[1][2]. The structural core consists of two RecA-like domains linked by conserved motifs that enable ATP binding and hydrolysis, as well as RNA binding, with variable N- and C-terminal domains conferring specificity[1][3]. Interacting drugs, mechanisms of action, and established biomarkers for DDX4 specifically are not firmly characterized in current published sources. However, the DEAD-box helicase family as a whole is an area of active investigation for oncology targets and biomarker development[1].
