Overview

DEAD-box helicase 54 (DDX54) is an ATP-dependent RNA helicase belonging to the DEAD-box protein family, characterized by the conserved Asp-Glu-Ala-Asp (DEAD) motif[1][5]. It participates in alteration of RNA secondary structure, including processes such as translation initiation, splicing, and ribosome and spliceosome assembly[1][2]. DDX54 plays a crucial role in ribosome biogenesis, particularly in remodeling rRNA secondary structures during 60S ribosomal subunit formation and is essential for establishing the peptidyl transferase center in ribosomes[2]. The protein can also repress the transcriptional activity of nuclear receptors, functioning as a transcriptional corepressor[3][5]. DDX54 is implicated in cellular processes related to cell proliferation, differentiation, and apoptosis, and its dysfunction is associated with diseases such as renal hypoplasia, duodenum adenoma, and potentially cancer[1]. There is currently no evidence for direct drug interactions or use as a specific biomarker[1].
