Overview

DEAH-box helicase 35 (DHX35) is a member of the DEAH (Asp-Glu-Ala-His)-box protein family, which are putative ATP-dependent RNA helicases characterized by conserved motifs involved in ATP binding and hydrolysis[3][7]. Like other DEAH-box proteins, DHX35 is implicated in numerous cellular processes that require alterations in RNA secondary structure, including translation initiation, pre-mRNA nuclear and mitochondrial splicing, and assembly of ribosomes and spliceosomes[7]. Functional studies have identified DHX35 as a regulatory factor in spliceosome complexes, acting in cooperation with the G-patch protein GPATCH1 to promote splicing fidelity and suppress cryptic splice site usage[3]. DHX35 has also emerged as a co-sensor with RIG-I, participating in innate immune responses to viral RNA by enhancing interferon and cytokine signaling in airway epithelial cells[1]. Although genome-wide association studies have linked DHX35 variants to increased susceptibility to certain types of pancreatic cancer, the precise molecular mechanisms underlying its role in disease remain to be fully established[3]. There are currently no reported drugs directly targeting DHX35, nor established mechanisms of action for pharmacological modulation, biomarkers, or specific safety concerns related to the protein.
