Overview

Death domain-containing protein 1 (DTHD1) is a poorly characterized protein encoded by the DTHD1 gene located on chromosome 4p14. The protein contains a canonical death domain, a protein-protein interaction domain found in many proteins that mediate apoptosis and intracellular signaling complex formation. Although death domain-containing proteins often participate in signal transduction, apoptosis, and inflammatory pathways, there is currently no specific evidence that DTHD1 is a validated therapeutic target or that it directly interacts with drugs or established cell signaling networks. Alternative splicing produces multiple transcript variants. In disease, DTHD1 has been associated in some reports with aniridia 1 and Leber Plus disease, but the connection is not well established and overall evidence for its direct involvement in human pathology remains limited. The broader protein superfamily (death domain proteins) is established as functionally important in apoptosis, inflammation, and immune response, but DTHD1 itself has not been directly linked to these processes in detail.
