Overview

Death domain-containing protein CRADD (CRADD) is an adaptor protein that contains a CARD (caspase recruitment domain) and a death domain, enabling it to interact with and recruit caspase-2 to cell death signaling complexes. Through its interactions, particularly with receptor-interacting protein kinase 1 (RIPK1) and tumor necrosis factor receptor 1 (TNFR1A), CRADD participates in the formation of signaling complexes—such as the PIDDosome—that lead to caspase-2 activation and apoptosis. Mutations in the CRADD gene are linked to certain neurodevelopmental disorders. While CRADD is essential in the apoptotic signaling pathway, it is not currently a direct therapeutic target for any approved drugs, but its key role in cell death makes it relevant in research on cancer and neurodevelopmental diseases[1][2][3][7].
