Overview

Death effector domain-containing protein (DEDD) is a ubiquitously expressed, predominantly nuclear protein characterized by an N-terminal death effector domain (DED) and multiple nuclear localization signals. It mediates nuclear and cytoplasmic signaling related to apoptosis, mitotic progression, and tumor suppression. DEDD can bind DNA and nucleosomes, inhibit rRNA transcription in the nucleolus, and regulate the Cdk1/cyclin B1 complex to modulate the cell cycle. It interacts with other DED-containing proteins, such as FADD and caspases-8 and -10, inducing weak apoptosis via caspase activation and helping shuttle apoptotic signals to the nucleus. In cancer cells, DEDD attenuates EMT and tumor metastasis partly through stimulating autophagy by modulating the PI3KC3/Beclin1 complex, leading to degradation of EMT regulators. Overall, DEDD integrates cell death, cell cycle control, autophagy, and tumor suppressive pathways but is not currently a direct therapeutic target[2][3][4][6][7].
