Overview

Death effector domain containing protein 2 (DEDD2) is a nuclear-localized protein characterized by the presence of a death effector domain (DED), which is a protein interaction motif crucial for regulating apoptosis and other cellular signaling pathways[3][4]. DEDD2 may regulate the trafficking of caspases—especially CASP8 and CASP10—and other proteins into the nucleus during death receptor-induced apoptosis, as well as contribute to the degradation of intermediate filaments and repression of polymerase II-dependent transcription[3][1]. As a homolog of DEDD, DEDD2 has about 48.5% sequence identity with DEDD and can interact with c-FLIP (an apoptosis regulator), DEDD, and is reported to play a critical role in the repression of gene transcription during apoptosis[1][3]. Disease relevance includes an association with colorectal adenocarcinoma[3]. DEDD2 is one of several human proteins containing DEDs, a family of apoptosis and signal transduction regulators essential for embryonic development and immune homeostasis[4]. No specific drugs, biomarkers for patient selection, or safety concerns have been described in current literature.
