Overview

Death Receptor 3 (DR3), also known as TNFRSF25, is a type II transmembrane protein belonging to the tumor necrosis factor receptor superfamily. It primarily mediates apoptosis and necroptosis, but more recognized functions are related to immune modulation and the regulation of T cell homeostasis, influencing inflammation and autoimmunity. Its ligand is TL1A. DR3 expression is highest in lymphoid tissues, including activated T cells and regulatory T cells.

Mechanism of action

TL1A binding initiates signaling cascades involving adaptor proteins like FADD/TRADD, leading to caspase activation (apoptosis) or RIPK1/RIPK3 activation (necroptosis). Can also activate NF-kB signaling.
