Overview

Death receptors are a subgroup of the tumor necrosis factor (TNF) receptor superfamily characterized by an intracellular "death domain" crucial for initiating apoptosis (programmed cell death) following ligand binding. Upon activation by their specific ligands (e.g., FasL for Fas/CD95, TRAIL for DR4/DR5), these receptors recruit adaptor proteins and caspases to form the death-inducing signaling complex (DISC), triggering a caspase cascade that results in cellular apoptosis. These receptors are major regulators of immune function and tissue homeostasis, and defects in their signaling contribute to cancer, autoimmune disorders, and impaired immune responses. Therapeutically, targeting death receptors (especially TRAIL-R1/DR4 and TRAIL-R2/DR5) has been pursued for cancer treatment due to their ability to selectively induce tumor cell apoptosis, although clinical success has been limited by challenges with biologic delivery, resistance, and insufficient apoptotic signaling strength.
