Dedicator of cytokinesis protein 11 (DOCK11) is a large intracellular signaling protein (~240 kDa) that functions as a guanine nucleotide exchange factor (GEF) specific for the small G protein CDC42, promoting the exchange of GDP for GTP and thereby activating CDC42 signaling pathways[1][3][5]. DOCK11 is a member of the DOCK-D subfamily and contains DHR1 and DHR2 domains, mediating membrane attachment and catalytic GEF activity, respectively[1][4][5]. It is expresssed predominantly in hematopoietic tissues and is critical for immune cell development (especially B cells), actin cytoskeleton regulation, and DNA repair processes, including those that maintain hepatitis B virus covalently closed circular DNA (cccDNA) in infected hepatocytes[1][3][5]. DOCK11 deficiency in humans causes early-onset and severe autoimmunity, while overactivity or dysregulation is implicated in viral persistence, fibrosis, and possibly pathological cell migration and cancer[1][3][5]. Emerging research suggests that DOCK11 is a promising therapeutic target for chronic hepatitis B infection through direct inhibition of its activity[1].
