Overview

Dedicator of cytokinesis protein 2 (DOCK2) is a large cytosolic protein and a member of the DOCK family of guanine nucleotide exchange factors (GEFs), specifically in the DOCK-A subfamily. DOCK2 is predominantly expressed in immune and hematopoietic cells, where it plays a central role in activating the small GTPase Rac, regulating actin cytoskeleton dynamics necessary for cell migration, adhesion, proliferation, and immune synapse formation. It orchestrates homeostasis and functional responses of various immune cells, including T and B lymphocytes, neutrophils, and dendritic cells, influencing chemotaxis and immune activation. Beyond immunity, DOCK2 regulates platelet development and function and has been implicated in disorders of hemostasis, inflammation, immunodeficiency, viral pathogenesis (including COVID-19 severity), cancer progression (via mutation or altered expression), and neuroinflammation. Pharmacological inhibition or genetic loss of DOCK2 impairs immune responses and platelet function, establishing it as both a research and emerging therapeutic target in autoimmunity, thrombosis, and inflammatory diseases.
