DCUN1D1 is a key component of the neddylation pathway, essential for activating cullin-RING ligases (CRLs) which regulate protein degradation via ubiquitination. It acts as an adaptor or scaffold that brings together components necessary for efficient neddylation, promoting transfer of NEDD8 from UBE2M-NEDD8 onto specific lysine residues on target cullins within CRLs. The *DCUN1D* gene is amplified in several cancer types—most notably squamous cell carcinomas—and may act as an oncogenic driver.
Facilitates NEDD8 transfer to cullins within CRLs, relieving CAND1-mediated inhibition.
