Defective in cullin neddylation 1 domain containing 4 (DCUN1D4) encodes a nuclear protein that serves as an adaptor/regulator in the ubiquitin-proteasome system, enabling cullin family protein binding and facilitating the neddylation (conjugation of the ubiquitin-like molecule NEDD8) of cullin-RING E3 ubiquitin ligases, which are essential for targeted protein degradation[1][2][3][4]. Disease associations involve cancer—where a circular RNA subtype (circDCUN1D4) is downregulated in tumors, suggesting a tumor-suppressor function—as well as possible roles in bone metabolism (osteoporosis, via exosomal miRNA networks) and in regulating platelet viability during storage[2]. No approved therapeutics are known to directly target DCUN1D4, though its molecular networks and biomarkers are of increasing research interest.
