Overview

Defective in cullin neddylation 1 domain-containing protein 3 (DCUN1D3) is a member of the DCNL (Defective in cullin neddylation-like) protein family that facilitates the transfer of the ubiquitin-like protein NEDD8 onto cullin proteins, thereby regulating the activity of cullin-RING E3 ubiquitin ligases such as the SCF (SKP1–CUL1–FBOX) complex[1][2][3][6]. DCUN1D3 plays a role in cell cycle progression, especially after UV-induced DNA damage, by modulating p27 protein degradation through the regulation of cullin neddylation and subsequent SCF^SKP2^ E3 ligase activity[1][2]. Knockdown of DCUN1D3 can block S phase progression and result in increased levels of the cell cycle inhibitor p27, implicating a function in cell growth and survival and suggesting possible tumor suppressor or cancer-related roles[1][2][5]. The protein localizes to both the cytoplasm and nucleus and its expression is induced by UV damage[1][2][3][5]. No direct therapeutic targeting, drugs, or use as a biomarker have been established in the current literature[2][5][6]. DCUN1D3 is involved in cullin neddylation and SCF ubiquitin ligase regulation, functions key to cell cycle control and DNA damage response[1][2][3][5][6]. It is not a classical therapeutic target (e.g., receptor, enzyme, transporter); rather, it is an E3 ligase-associated cofactor and neddylation regulator[1][2][3]. No drugs are currently known to directly interact with DCUN1D3, and there are no established mechanisms of action relevant to drug targeting, biomarker roles, or safety concerns according to available references[2][5][6].
