Overview

Defensin beta 122 (DEFB122) is a member of the beta-defensin family, a group of small, cationic, disulfide-rich antimicrobial peptides vital to innate immunity and primarily expressed in epithelial and reproductive tissues[1][3][5]. DEFB122 is located within the human beta-defensin cluster on chromosome 20q11.1, which comprises several genes predominantly expressed in the male reproductive tract and implicated in both antimicrobial defense and reproductive function[1][3][5]. However, human DEFB122 is reported as a pseudogene (DEFB122P), meaning the gene is not believed to encode a functional protein due to a disruption in the coding sequence which leads to truncation and loss of the conserved defensin structure[1]. Thus, while functional orthologs exist in macaques, the human DEFB122 gene does not produce a functional protein and is not considered an active therapeutic or diagnostic target[1]. The human DEFB122 gene is a pseudogene and, as such, does not produce a functional peptide with the canonical defensin fold due to a shifted splice site and premature stop codon[1]. Beta-defensins in general play roles in host defense, particularly antimicrobial activity against bacteria, viruses, and fungi, as well as involvement in fertility and sperm function, but these roles pertain to other members of the family and not to human DEFB122 itself[1][2][3][5].
